学术报告:用超分辨率成像方法解析体细胞和干细胞中三维基因组和环状结构

报告摘要:

Dissecting the 3D-chromatin organization in cell physiology is a key area of investigation. By using quantitative super-resolution nanoscopy, we identified a novel chromatin fiber assembly and its relation with naïve pluripotency. Nucleosomes are arranged in groups of various sizes, the nucleosome clutches, which control gene function. We recently visualized the structure of cohesin-mediated loops in human cells and discovered that transcriptional-dependent supercoiling controls loop formation and 3D-genome organization. Furthermore, by combining imaging and genomic approaches, we designed MiOS, a powerful integrated strategy to model the folding of key pluripotency genes at nucleosome resolution. Overall, super-resolution microscopy combined with genomic and modeling methods allowed us to dissect the functional role of transcription-mediated supercoiling and the nucleosome level structure of genes, which ultimately are key features controlling gene activity.

Maria Pia Cosma received the PhD in Cellular and Molecular Genetics, School of Medicine, Univ. of Federico II in the year 2000. From 1997-2000 she was Marie Curie Post-doc at IMP, Vienna and from 2000 to 2003 she completed her education at Telethon Institute of Genetics  and Medicine (TIGEM), Naples. By the end of 2003 she established her first group at TIGEM  in Naples and become also EMBO Young Investigator (YIP). From 2004 to 2010 she has been  lecturer at SEMM (European School of Molecular Medicine) and Honorary Associate  Investigator at IGB, CNR, Naples from 2009 to 2015. In April 2010 she moved to Barcelona,  Spain, at the Center for Genomic Regulation (CRG), as Senior Scientist and ICREA Research  Professor. She became co-coordinator of the Systems and Synthetic Biology Program at CRG  since February 2023. At the end of 2018 as 1000 Talent Awardee, she joined the Guangzhou  Institute of Bio-medicine and Health (GIBH), Guangzhou, China, and she is currently working  at Guangdong Provincial People’s Hospital (GDPH), Guangzhou, China. She has given many  invited talks at major international conferences and Research Institutes. She received several  prizes including: Marie Curie Excellence Award in 2005, the honour of Order of Merit of the  Italian Republic in 2007, La Vanguardia de la ciencia 2014, Barcelona City Prize in 2015, La  Caixa Health 2018. She is ERC stGrant awardee, 2009 and HFSP Grant awardee, 2010. She  has been elected EMBO Member in 2010. She has been the coordinator of the FET-Open  project CellViewer and of the associated FET-launchpad project. She is currently the  coordinator of the EIC Pathfinder project EcaBox. She was elected EIC National Champion in 2021 and EIC Woman Leadership Programme awardee in 2022.

Cosma’s group has the main objective to investigate the controlling mechanisms of stem cell  maintenance, the reprogramming of somatic cells, and tissue regeneration. (Lluis et al., Cell  Stem Cell 2008; PNAS 2011; Sanges et al., Cell Reports 2013; J. of Clinical Investigation 2016;  Pedone et al., Cell Reports 2017; Frade et al., Science Advances 2019, Aulicino et al. Stem Cell  Reports 2020, Arumugam et al Cell Reports 2020, Pesaresi et al. Molecular Therapy 2021).  Cosma’s Group has a long interest in studying gene and chromatin function (Cantone et al.,  Cell 2009). Recently using super resolution microscopy (STORM), the group has identified a  new model of chromatin fiber assembly and how the fiber changes in somatic and embryonic  stem cells (Ricci et al., Cell 2015; Neguembor et al., NAR 2018; Otterstorm et al., NAR 2019;  Cosma & Lakadamyali, Nature Methods 2020; Neguembor et al. Molecular Cell 2021;  Castells-Garcia A et al. NAR 2022; Neguembor et al. Nature SMB 2022).
